no, not while it is running. quickest way to ruin a USB portable hard drive (unless thumb or pen drive) is to smack it around while it's spinning. Replaced plenty of those to my customers who "tripped" or snagged the cable and lost their entire backup..........

For a bulletproof backup solution, get an external hard drive, cd's or DVD's and backup daily, or hourly depending on how critical your work is. Large Database operations or accounting done by multiple peeps on a server is one good example of daily multiple backups.
